A separate stabbing attack was reported near the former Israeli settlement of Ganim, where Israel has recently established a military base.
The Israeli military said soldiers killed a Palestinian who stabbed a soldier in the leg near Ganim.
The Palestinian health ministry identified the three Palestinians killed as Mahmoud Abu Samra, 20, Mohammed Militat, 16, and Yazan Sawafteh, 25.
A Magen David Adom medic said the Israeli civilian wounded near Itamar was stabbed in the chest and taken to hospital in serious condition.
A Palestinian municipal official said Israeli settlers lit the fire near Itamar and that Israeli soldiers opened fire after residents went to extinguish it.
Israeli police and fire services are investigating the cause of the fire near Itamar.
